Does my probe sequence need to be modified to convert it to a ZEN double-quenched probe?

The sequence does not need to be altered to accommodate an internal ZEN modification. IDT has tested many assays with direct substitution of ZEN/IBFQ into the probe sequence and has found no performance issues.

Note that when ordering a ZEN modification, both the assay and probe ordering tools will automatically insert the ZEN quencher in the appropriate location.

Read more about the uses of the ZEN secondary quencher at Modification Highlight: ZEN™ Internal Quencher.
